Regular autophagy activation is essential for cellular health. Spermidine, a naturally occurring polyamine, has been theorized to promote autophagy, which helps reduce inflammation, improve lipid metabolism, and regulate cell growth and death. Both spermidine supplementation and intermittent fasting are powerful autophagy inducers, optimizing lipid metabolism and reducing inflammation, particularly the age-related kind known as inflammaging. • As we age, autophagy declines, partly due to a reduction in spermidine levels. This is particularly evident in immune cells and other tissues. Spermidine's promotion of mitophagy—the degradation of damaged mitochondria—supports mitochondrial health, enhancing energy efficiency and reducing oxidative stress, a key factor in aging. Additionally, spermidine upregulates heat shock proteins, offering cells protection against damage, similar to the effects of fasting. • Both spermidine and fasting activate longevity pathways such as AMPK and mTOR. Overactivity of mTOR, a nutrient-sensing pathway, accelerates aging, but fasting and spermidine inhibit it, amplifying their longevity-promoting effects. Increased endogenous spermidine levels enhance the anti-aging benefits of fasting. Spermidine supplementation is linked to increased survival and extended health span in humans and animals. Epidemiological studies also support the association between higher dietary spermidine intake and reduced mortality. • Spermidine and fasting both promote cardiovascular health by reducing blood pressure and protecting the heart. Spermidine delays cardiovascular disease onset, preserves diastolic function, and reduces cardiac hypertrophy in animal studies. High dietary spermidine intake correlates with lower cardiovascular disease incidence in humans. • Spermidine may also lower cancer-specific mortality and has potential as a chemopreventive agent. As spermidine levels decline with age, centenarians often have higher levels of spermidine and spermine, another polyamine linked to longevity. Supplementation with spermidine has been shown to be safe and well-tolerated, especially in older adults, with studies indicating modest improvements in memory. • Dietary sources of spermidine include matured cheeses, mushrooms, organ meats, and red meat. Cooking methods like boiling or grilling can reduce polyamine levels, while techniques such as sous vide or microwaving preserve them. Mushrooms, besides being rich in spermidine, contain other bioactive compounds like chitin. • Chitin, and its derivative chitosan, has various health benefits, including improved glucose tolerance, increased insulin secretion, and protection of the gut epithelium. Chitosan also has antiviral, anticancer, antifungal, and antibacterial properties, contributing to overall health and longevity. Including prebiotics like chitin in a ketogenic diet could have synergistic effects for radical longevity extension.
