DNA isolation from fish

This is a practical video about DNA isolation from fish performed by bio-technologists. • While sampling according to the requirement weigh the sample, here we have taken 2 gm of fish fin tissue sample. You can store the sample by adding 95% ethanol or can use fresh sample as like ours. • Add lysis buffer 500µl (Tris HCL, EDTA, SDS) and homogenize until fine crushed. Add proteinase k 2µl and vortex the sample. Incubate the sample overnight. • Add equal amount of Phenol chloroform isoamyl-alcohol (25:24:1) and mix it thoroughly by inversion for 5mins. • The sample was centrifuged at 10000rpm for 10 min at 27 °C, and the supernatant was placed into a new, sterile mirco-centrifuge tube. • Repeat the PCI step and centrifuge it again at 10,000rpm for 10 min. • Transfer the supernatant into fresh tube and add absolute ethanol, centrifuge 10000rpm for 10 min at 4 °C. • Discard the supernatant and air dry the pellet. • Add 30µl of Nuclease free water to dissolve the pellet and store at -20°C. • Run the samples on 0.8% agarose gel for electrophoresis for 45min at 80 volts. • After electrophoresis you can visualize the gel under gel documentation unit. • Amplicons/bands of DNA observed, confirms the presence of isolated DNA. • To watch our more videos click on the link below- • • National Education Policy 2020:भारत क... • • How bacteria's can talk with each oth... • • What happens when plants are exposed ... • Follow us on instagram- / amplicons_biotech • #Biotechnology #DNA isolation #Fish DNA isolation #Extraction of DNA #DNA #Molecular biology #Genetics #Genetic engineering #science #Method of DNA isolation
