Can hand sanitizer with 70% ethanol kill bacteria? This short video shows real-time killing of E. coli with 70% ethanol. The first 6 seconds show live bacteria. Application of a few drops of ethanol instantly kills all bacteria indicated by the absence of movement. • At Danish Technological Institute (DTI), we help importers and producers of antimicrobial products document the efficiency of their products. (Danish) • Bacteria: Escherichia coli. • Samples: Micropore filter biofilm prepared by a drop of overnight bacteria onto a 0.22 µm polycarbonate filter placed on nutrient agar plate. The plate was incubated at 37 °C for 2 hours before microscopy. • Microscopy: Spinning disk microscope. • Recording: Time lapse imaging was performed on an Olympus IX83 inverted microscope using bright-field illumination and a 100x 1.49 NA TIRF objective. The acquisitions were obtained with a Hamamatsu ORCA-Flash4.0 sCMOS camera, and the system was controlled by Olympus CellSens software. Image analysis was performed in Olympus CellSens and Fiji/Image.
