Hyperprogression is an increased cancer growth rate caused by immune checkpoint inhibitors (PD-1/PD-L1) like Opdivo and Keytruda, occurring in 4-29% of treated patients. It differs from pseudoprogression, as hyperprogression leads to a significant increase in tumor size and new tumor locations, reducing patient lifespan. Studies suggest a link to MDM2 gene amplification, EGFR aberrations, and tumor-associated macrophages (TAMs). Removing part of the antibody that binds to TAMs can prevent hyperprogression. Drugs like clodronate, CSF-1R inhibitors, Anti-MARCO antibodies, and natural substances like 6-Gingerol and Silibin may help counter TAMs. Immunotherapy can cause serious side effects, requiring patient awareness and monitoring. • Reference: Jason R.
