Activity 2. Genomic DNA Extraction using Phenol/Chloroform (Liquid-Liquid Extraction Method) • Conventional method of genomic DNA (gDNA) extraction using phenol/chloroform utilize detergents such as sodium dodecyl sulfate (SDS) or cetyltrimethylammonium bromide (CTAB) for cell lysis. After cell lysis, phenol/chloroform/isoamyl alcohol (25:24:1) is added to separate lipids and other proteins into the organic phase, and the gDNA into the aqueous phase. After centrifugation, the aqueous phase containing the gDNA is transferred to a clean tube for precipitation using sodium acetate and ethanol. Precipitated gDNA is pelleted by centrifugation and eluted using Tris-EDTA buffer. Phenol/chloroform DNA extraction method can be used using various samples including animal blood samples, plant tissues, fungal samples, and bacterial cells. • #dna • #molecularbiology • #biotechnology
