Mouse macrophages phagocytosing apoptotic human cervical cancer HeLa cell Version 2

Apoptosis was induced in human cervical adenocarcinoma cells (Hela cells, ATCC, CCL-2) by UV exposure in DPBS (Gibco, Cat# 14190-144 http://bit.ly/VhzUTs) for 5 hours. DPBS supernatant was saved. Apoptotic Hela cells were labeled with pHrodo™ Red, SE (Life Technologies, Cat# P36600 http://bit.ly/VsOGJi). Mouse monocyte-macrophage cells (MMM cells, ATCC, Cat# TIB-67) were plated on 35mm MatTek dishes at a density of 250,000 cells per plate. Labeled MMM cells with Hoechst 33342 (Life Technologies, Cat# H3570 http://bit.ly/SHPkBJ). Fed 300,000 of the apoptotic, pHrodo™ Red labeled Hela cells to one plate of the Hoechst 33342 labeled MMM cells in a 1:1 solution of Live Cell Imaging Solution (Life Technologies, Cat# A14291DJ http://bit.ly/UCBuTp) and DPBS supernatant from UV treatment. Imaged on Applied Precision Delta Vision system with Olympus IX71 Microscope and Photometrics CoolSnap HQ2 Camera. Time-laspe imaging every 45 seconds for 5 hours. 40x. Channels: DIC, Dapi, and TRITC. Deconvolved and Zoomed. Movie speed: 10 frames per second for 1 and 7.5 frames per second for 2.
