Hello everyone • Microscopes you might know from biology classes in school, are usually #wide-field microscopes (TechThursday XLII). This means that the entire sample you want to measure is illuminated at once and all the signal reaches the detecter unfiltered, including the background signal which in turn leads to lower contrasts in the pictures. • Today we want to show you a different type of #microscope: the Confocal Laser Scanning Microscope (CLSM) which allows us to increase the resolution and the contrast significantly. In this case a point #illumination is applied to a very small area and a pin-hole is placed in front of the detector to filter out any light which is not in focus (therefore confocal). Like this a very high optical resolution can be achieved and also the contrast is much better because most of the background signal is excluded. • Using this technique alone, only a very small part of the sample can be observed. Thus, to get a complete picture, a bigger area needs to be scanned and the generated pictures merged. Since the focus point of the light can be changed in all 3 dimensions, using the scanning technique 2D- or even 3D-images can be generated, and even the movement of e.g a droplet in solution can be observed… pretty amazing, isn’t it? • Have a wonderful Thursday!👋 • #NCCR #NCCRmse #Science #CLSM #confocal #microscopy
