this is the video lecture regarded to... • • in this video lecture we will explain.... • in this video lecture we will explain the following content related to this lecture • 1. introduction • PDA is a general purpose or basal growth media for fungi. • Chloramphenicol, Tartaric Acid and Chlortetracycline can be added as selective agents to inhibit bacterial growth • Recommended by APHA and FDA for plate counts of yeasts and molds in the examination of foods and dairy products. • 2. composition • 3. Uses • Used for the detection of yeasts and molds in dairy products, prepared foods and also from clinical specimens. • PDA with Chloramphenicol is recommended for the selective cultivation of fungi from mixed samples. • PDA with Chlortetracycline is recommended for the microbial enumeration of yeast and mold from cosmetics. • It also aids in cultivating and differentiating pathogenic and non-pathogenic fungi. • PDA is also useful for maintaining stock cultures of certain dermatophytes. • 4. Principle • • Dextrose act as a carbohydrate source which serves as a growth stimulant • Potato infusion provides a nutrient base for the luxuriant growth of most fungi. • Agar is added as the solidifying agent. • The addition of supplement like chloramphenicol (25 mg), Chlortetracycline (40 mg) and tartaric acid (1.4 gm) inhibit bacterial growth. • Distill water mix all the ingredients. • 5. Preparation • Suspend 39 grams in 1000 ml distilled water. • Heat to boiling to dissolve the medium completely. • Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. • Cool the medium to approximately 50°C and pour into sterile Petri dishes. • Inoculate and streak the medium with one isolated pure colony. • Incubate in an aerobic atmosphere at 35ºC for 24-48 hours. • Observe the growth of fungus. • 7.
