They are accountable for destroying germs in injured or infected tissue. • Eosinophils: Eosinophils treat infections caused by parasites and also control the immune system's response to allergic reactions. • Basophils: These are the least common type of white blood cells. Their function is unknown, but they may play a role in allergic reactions. • 2.) #Agranulocytes: • Lymphocytes: There are three types of lymphocytes. • @ B lymphocytes: produce antibodies to attack bacteria, viruses, and other foreign invaders. • @ T lymphocytes: help to identify cells that require an immune response. • @ Natural killer cells: destroy cancer cells and viruses. • Monocytes: These cells are similar to neutrophils. They destroy bacteria, those causing chronic infection and also repair damaged tissues. • The function of each type of WBC is specific and their number is also different per 100 counts. • #Neutrophils: 40–60 percent • #Eosinophils: 1–4 percent • #Basophils: 0.5–1.0 percent • #Lymphocytes: 20–40 percent • #Monocytes: 2–8 percent • • Different physiological and clinical conditions cause variation in different WBC count ( DLC Test ) • Hence the deviation from the normal count of different WBCs per 100 WBC helps to detect is Eosinophilia, tuberculosis, malaria and other diseases • This type of counting is known as differential count. • Let's begin with our practical DLC test: • • To stain WBCs, we are going to use Leishman stain that contains both acidic and basic dyes • Blood Film Preparation: for DLC test • Sterilize your fingertip with rectified spirit and pri.k it with needle • Place a drop of blood at one end of the clean slide. • Use another slide as a spreader to spread that blood drop into a thin smear. • To achieve a thin layer of blood, maintain a 45-degree angle between spreader and blood slide. • You will get uniform blood film in a tongue shape nature. • Till the slides get dried, prepare the staining rack. • • Put the air-dried slide on the staining rack and add sufficient amount of Stain with the help of dropper on the blood film. • Wait for 1 to 2 minutes to allow proper fixing. • After 2 minutes add the double amount of distilled water or Leishman buffer over the film carefully. • After 10 minutes wash the slide under running water. • Let the slide get dried in the air. • Start the counting from one end of the film to another and note the different types of leukocytes from each field under the Microscope. • The counting is continued till the total number of leukocytes i.e. WBCs become hundred. • • Previous video: WHITE BLOOD CELLS • • White Blood Cell Identification | WBC...
