Progeria • Progeria is caused by a mutation in the LMNA gene that produces the Lamin A protein. Researchers still do not know the exact cellular events that lead to the process of premature aging seen in Progeria. Progerin, which is a truncated version of abnormally processed Lamin A protein caused by the mutation, is thought to affect the structure and organization of the nucleus, thereby causing changes in nuclear mechanics and affecting gene regulation, chromatin organization, and DNA repair, leading to the premature aging. • Children with Progeria begin to display many characteristics of accelerated aging at around 18-24 months of age. Progeria signs include growth failure, loss of body fat and hThe Progeria Research Foundation, Inc. (PRF) is the only non-profit organization dedicated to Progeria research. PRF has also hosted numerous workshops to allow scientists and clinicians to come together and share ideas on Progeria pathophysiology, treatment, and cure. • In this video clip, the President and Executive Director of PRF, Audrey Gordon talks about the role that progerin plays in helping us understand both the pathophysiology of Progeria but also of the aging process and cardiovascular disease. • Progeria, which is caused by a mutation in the LMNA gene that produces the Lamin A protein, have increased levels of progerin. Researchers believe that progerin affects the structure and organization of the nucleus, thereby causing changes in nuclear mechanics and affecting gene regulation, chromatin organization, and DNA repair, leading to the premature aging. That concept may also exist in the normal aging process and as such, progeria research may lead to a better understanding of aging and cardiovascular disease. • At this year's PRF Scientific Workshop being held in Bethesda MD, April 24-216, 2013, there will be several studies presented on aging and cardiovascular disease.
