Touchdown PCR - This lecture explains about the mechanism and principle of touchdown PCR reaction and the application of touchdown PCR in amplifying DNA fragments. Touchdown PCR is done to eliminate the formation of unwanted DNA amplification and it increases the specificity of the PCR reaction process. In touchdown PCR, we start with a very high annealing temperature which is close to the melting temperature of the primer and slowly reduce the temperature as PCR cycles are conducted.
