100 vs 125 grain broadhead and does it matter

I've always wondered whether I should shoot 125 up front or 100. I've normally went with 100 just because that's the normal grain to go with and I shoot a shorter draw and 60 pounds. I wanted to do some testing in the back yard and see how big of a difference at 20,30,40 100vs125 would make and whether or not it would make an arrow flex more at each distance even changing from 340 to 400 spine arrows.
